Abstract
The complete mRNA sequence of a specific gene in tobacco (Nicotiana tabacum), known as 2-hydroxyisoflavanone dehydratase, was amplified using rapid amplification of cDNA ends (RACE) methods. The mRNA produced by the tobacco 2-hydroxyisoflavanone dehydratasedase gene was determined to have a length of 1,278bp, and it contains an open reading frame of 966 bp that codes for a protein consisting of 321 amino acids. After conducting sequence analysis, it was found that the tobacco 2-hydroxyisoflavanone dehydratasedase exhibits significant similarity to comparable genes in other plant species, including nicotiana tomentosiformis (99%), capsicum annuum (78%), potato (75%), lycopersicon pennellii (73%) and lycopersicon esculentum (72%). Upon further analysis of the tobacco genomic sequences database, it was discovered that this specific gene does not contain any introns and is composed of a single exon. Furthermore, our results suggest a closer genetic relationship between the 2-hydroxyisoflavanone dehydratasedase gene in tobacco and its equivalent in nicotiana tomentosiformis. Furthermore, tissue expression profile analysis demonstrated high expression levels of this gene in both flower and leaf tissues, while moderate expression was noted in both the root and stem tissues. This experiment serves as an important foundation for future investigations into the functions and characteristics of this specific gene in tobacco.
